Mmm. We thought of having a letter column on our comic, but we ended up having comments instead; it felt kind of overkill to have both.
We keep it kinda minimal. There's an "about" page, which both teases the story and has super-brief character bios. An "archive" with tiny thumbnails of the whole story. A fan art gallery, since we had a little of that before the strip even launched; links to stuff we like, a page with the t-shirts and book, and links to the RSS and the Livejournal syndicated feed.
The less the better, I say. The site's there to showcase the irregular flow of pages. Everything else is ancillary.
I agree with Egypt...simple is best. The more you add, the more you have to manage and update and I'd think the comic is enough. I really enjoy your blog updates and reading others' comments. Beyond that, I'd just keep it to things that you're really inspired to do versus adding bells and whistles for your readers, imho. :)

The characters page has some display issues ... some left side cutoff when viewed in IE8 at 1280 x 1024.
As far as the design, I agree with Egypt and Nicole ... simple and clean is nicest. Make sure that your communication paths are few so that you have fewer places for people to have to search to get your news.
